| A series of primary studies were conducted in this research project, in order to determine the appropriate sterilization method, number of nodes, appropriate sampling period of explants and growth medium and also to induce shoot growth and prevent browning between 1984-1993 years, so as to achieve guick sapling production by tissue culture method.
Explants those are the nodes of annual shoots were used as material in this study.
At the end of a series of researches it was concluded that the best result is obtained on the condition that third and fourth nodes being kept 10 minutes in Ascorbic acid solution of 10 mgl, then being sterilized by dipping in 50-70 % Ethanol for 5-15 seconds and by soaking in Sodium hypochloride of 10% for 15 minutes; later being cultivated at 5.8 pH, including 0.5 mgl-1 BAPon OM( Olive Medium) ; on the further weeks containing 3 gl-1 active charcoal.
Besides, keeping the explants in 50 mg/l Gibberellic acid solution for shoot elongation and 0.5 mg/l BAP addition in the growth medium for shoot formation gave positive response. The best sampling period of explants were decided to be June-October for Gemlik cv. And August-September for Memecik, Ayvalık cv. |
