| In this study, in order to get use of somaclonal variations which appear during the callus culture of olive, the effect of the factors like different hormone combinations and concentrations, explants, periods, photoperiod and darknes on callus formation, were investigated.
The best callus growth was obtained in OM containing 1 mg/ l NAA+0.5 mg / l BAP , under the photoperiod.
Ayvalık variety gave better results than Gemlik in callus formation, and the best callus growth was obtained in petiols in December and January.
The best shoot growth from one year old shoots of Gemlik was obtained on the media supplemented with 4 mg/l Zr in April and May. Leaves taken from in vitro grown shoots gave highest callus formation in 1 mg/l IBA + 0.5 mg/l BAP combination , but regeneration wasn't observed. Regeneration was achieved from the internodes of in vitro grown shoots on medium containing 4 mg/l Zr + 1 mg/l IBA. |
